Home Assignment – III (Activity)

Activity-2

Aim: -To prepare a temporary slide of onion peel.

Materials Required:

Onion peel, forceps, dropper, safranin solution, glycerin, microscope, Distilled water,
needle and brush, cover slip, glass slide and watch glasses.

How to proceed :

1. Take one onion bulb and remove it’s dry pink outer coverings.
2. Separate the fleshy coverings.
3. Remove the thin layer, keep it on a slide and put a drop of water on it to
prevent it from drying.
4. Cut it into a small piece and put a drop of safranin solution to it.
5. Put a drop of Glycerin and place a coverslip by using the needle. Glycerin
keeps the cells hydrated.
6. Observe it under the microscope.
